CLS Cell Lines Service bietet menschliches Thrombozytenlysat (hPL) an, einen der am häufigsten verwendeten Ersatzstoffe für fetales Rinderserum (FBS) in der Kultur von menschliche Zelllinien. Bei ersten Versuchen wurde es verwendet, um das Wachstum von mesenchymalen Stromazellen zu unterstützen.
Anschließend wurde hPL für die Vermehrung verschiedener menschlicher Zelllinien getestet und zeigte eine gute Wachstumseffizienz. CLS konnte zeigen, dass die wachstumsfördernde Wirkung von hPL ähnlich zu FBS, in einigen Fällen sogar überlegen, ist. Wir haben die Vermehrung verschiedener Zelllinien ausgiebig getestet, um sicherzustellen, dass unsere hPL-Lösung eine sichere und zuverlässige Alternative zu FBS ist.


Herstellen
Humanes Thrombozytenlysat wird mittels Thrombozytenapherese unter Verwendung von Blut gesunder Spender aus zertifizierten Blutzentren hergestellt. Um chargenspezifische Unterschiede zu vermeiden, werden die menschlichen Blutplättchen in grossen Mengen zusammengefasst, um die Konsistenz zu gewährleisten und damit die Reproduzierbarkeit zu verbessern.

Alternativen
Neben hPL bieten wir auch vollständig serumfreie Lösungen für verschiedenste Zelllinien an. Unsere umfangreiche Forschung und Entwicklung führte zur Formulierung von SciNX und SciN4Vero. Diese wurden an einer Vielzahl von Zelllinien getestet und unterstützten das Wachstum und die Proliferation. Falls serumfrei Lösungen keine Option sind, ist hPL die beste Alternative zu FBS, um reproduzierbare und sichere Zellkulturbedingungen zu gewährleisten.

    Humanes Thromobozyten Lysat enthält zahlreiche Proteine und Wachstumsfaktoren (Zytokine) und stellt eine hervorragende Alternative zu Foetalem Rinderserum (FBS) dar. So unterstützt hPL nicht nur die Expansion von mesenchymalen Stromazellen (hMSC), sondern auch von unterschiedlichen Zelllinien. Wir von CLS haben erfolgreich einige unserer immortalen humanen Zelllinien (A-549; Caco-2, HaCaT, MCF-7, U-251 MG) sowie humane Gingiva-Fibroblasten (HGF) und Dentale Pulpa Stammzellen (DPSCs) kultiviert. Die Zugabe von 5% hPL anstelle von 10% FBS zu den entsprechenden Zellkulturmedien war in allen Fällen für eine gute Proliferation ausreichend.

    Dieses hPL wird im industriellen Maßstab in Anlehnung an GMP Standards hergestellt.

    DMEM (Dulbecco's modification of MEM) cell culture medium supplemented with hPL (human Platelate lysate)

    DMEM contains four-fold concentrations of the amino acids and vitamins as compared to BME (Basal Medium Eagle). This culture medium, enriched with 4.5 g/L glucose, supports the proliferation of a wide variety of normal and transformed cells, such as primary cultures of mouse and chicken cells growing adherently.

    The DMEM:Ham's F12 is a 1:1 mixture of DMEM containing 4.5 g/L glucose and Ham's F12. As this medium is rich in amino acids, hormones and minerals, it supports the growth of many cells and cell lines even under low serum conditions. Thus, it is possible to work with serum concentrations of 5% or less. Furthermore, it is an excellent basic medium for the serum free cell culture.

    The EMEM medium is one of the first chemically defined cell culture media developed in 1959 by H. Eagle. This medium is ideally suited for culturing a variety of normal and neoplastic cells and cell lines. For a couple of cell lines, the addition of NEAA (Non-Essential Amino Acids) and Pyruvate is recommended.

    IMDM medium is a chemically defined cell culture medium which contains additions of selenium, aminoacids and vitamins, sodium pyruvate and HEPES. It is ideally suited for culturing B lymphoblastoid cells and hybrid cells. The buffering capacity is based on HEPES and hydrogen carbonate.
